Antigen:

C4 is a component of the classical complement pathway and is split by activated C1s to C4a and C4b. Together C2a and C4b forms the classical C3 convertase (1). The molecular mass of C4 is 204 kDa and it consists of 3 subunits: a (97kDa), b (75 kDa) and g (33 kDa).

C4 isolated from human plasma

Specificity:

HYB 162-02 is specific for human C4b

Epitope specificity:

Epitope specificity differs from that of HYB 162-04, as determined by inhibition ELISA.

Reactivity:

HYB 162-02 reacts strongly with C4. A strong reaction is seen with normal human serum when tested in sandwich ELISA in combination with a polyclonal antibody against C4 (e.g. DAKO A065). No reaction is seen with C4-depleted serum. HYB 162-02 also reacts with C4b deposited on coated antibody molecules. In Western blotting after SDS-PAGE, HYB 162-02 reacts with C4 in both reduced and non-reduced forms.
